Does tamping pressure change the shot?

Far less than people assume, once the bed is sealed. Beyond the point where the puck is compacted, extra force does very little because coffee stops compressing. What matters is that the tamp is level, since a sloped puck gives the water a thinner path on one side. Level beats hard, every time.

What is channelling and how do you see it?

It is water finding a low resistance path through the puck rather than passing evenly through it, so part of the coffee is over extracted and the rest barely touched. The result tastes both bitter and sour. A bottomless portafilter shows it instantly as jets or sprays instead of a single even stream, which is why it is such a useful diagnostic tool.

Is a puck screen worth adding?

It helps in some setups by spreading the water arriving at the top of the puck and by keeping the shower screen clean. It is a smaller effect than distribution and it is not a fix for channelling caused by clumps. Cheap enough to try, not a substitute for stirring the bed.

Does the number of needles matter?
